LCMS Study of the Occurrence of Caffeine and Theobromine in Scalp Hair

1. Correlation of coffee consumption with caffeine concentration in hair between individuals does not appear to be comparable. 2. NaOH hydrolysis chemically alters caffeine. 3. Preliminary work indicates direct MeOH/HCl or CHCl_3 extraction can isolate caffeine and theobromine but further investigation is required. 4. Solubility issues of theobromine in CHCl_3 may have led to quantitation difficulties. 5. Spiked matrix indicates recoveries of 85%. Possibly due to caffeine distribution differences in scalp hair. 6. Calibration in matrix has good correlation with aqueous standard.
