摘要

The combination of AEC X HILIC and exoglycosidase digestion is a powerful approach for comprehensive characterization of the oligosaccharides present on complex glycoproteins such as EPO. The determination of up to 5 poly-N-acetyl lactosamine repeats is a novel finding. Although present at low levels, these glycans may infer additional functional interactions for the drug including recognition by galectins or galactose binding proteins resulting in either secondary biological activity or more facilitated clearance. However, these large oligosaccharides may also be beneficial, masking immunogenic conformational epitopes on the protein.
