Protein identification on intact biofilm was performed with minimum sample preparation. In situ tryptic digestion at spatially localized spots on intact biofilms was used for imaging proteomics. Used both top-down and bottom-up proteomic approachs to identify proteins on intact biofilms. Spatial localization of the identified proteins on intact biofilms were obtained by MALDI imaging. Subcellular localization of the identified proteins indicate most of them are associated with the cellular membrane. Spatial localization of these membrane proteins in the presence of antibiotics can aid novel antibiotic development.
