CENP-A confers the epigenetic element of centromeric chromatin identity. The molecular mechanisms for CENP-A localization and maintenance are poorly understood. We hypothesize that post-translational modification of CENP-A may be involved in these activities. To this end we have generated highly pure CENP-A samples from bacteria and human cells, and performed analyses using nanoflow microcapillary liquid chromatography, electron transfer dissociation (ETD) fragmentation, and high resolution MS and MS~(2) acquisition (nanoLC-ETD-MS/MS). We observe that CENP-A from human cells co-purifies with nucleic acid which we believe to confer protection from protease treatment.
