Rapid Assay Development and Refinement for Targeted Protein Quantitation Using an Intelligent SRM (iSRM) Workflow

The newly developed intelligent SRM (iSRM) technology provides the easiest and fastest way to develop the robust, accurate, and sensitive SRM assays for targeted protein quantification. A validated, highly complex iSRM assay to simultaneously quantify and identify 40 yeast proteins was developed in a few hours by using the Pinpoint software and the iSRM workflow. The analytical precision of the validated iSRM assay was excellent. Ninety-two percent of the targeted peptides had (percent)CVs <=5percent. Ninety-nine percent of the targeted peptides had (percent)CVs below 10percent and all the peptides had (percent)CVs below 12percent.
