摘要

Protein glycosylation is one of the most important post-translational modifications and plays crucial roles in diverse biological functions. However, it is still a great challenge to analyze the glycoproteins due to the facts that glycoproteins usually bear enormous structural complexity of glycans and present at a relatively low concentration. An efficient enrichment step is essential for characterization of glycoproteome prior to mass spectrometric analysis. To address this issue, we developed a nanoprobe-based affinity mass spectrometric method which integrates the newly designed hybrid lectin-phenylboronic acid functionalized magnetic nanoparticles (BA-lectin@MNPs) and direct protein identification by MALDI-TOF MS for rapid, versatile, and efficient detection of glycoproteins.
