Quantification of Prostate Cancer Proteins from Expressed Prostatic Secretion Using a Stable Isotope Labeled Proteome

A SILAP proteome standard specific to PCa was successfully created for global quantification of protein candidates in EPS samples from PCa patients. It has been demonstrated that the PC-3 SILAP standard can serve as an internal standard for simultaneous quantification of several protein targets in a single analysis. Ongoing studies are aimed at utilizing the PC-3 SILAP secretome standard for high throughput selected reaction monitoring (SRM) of select proteins from a large cohort of EPS from PCa and BPH patients. Differentially quantified PCa-specific EPS proteins will be validated in a prospectively gathered blind study set of men with PCa and BPH by SRM.
