An ANP method using the Diamond Hydride stationary phase provided a highly robust untargeted metabolite profiling of E. faecium (EF) and S. aureus (SA). With the ANP method used it was shown that this approach can differentiate bacteria according to phylogeny and antibiotic susceptibility. These results demonstrate that ANP chromatography is a robust tool for pursuing untargeted metabolomicsbased studies of bacteria such as E. faecium and S. aureus. Three main classes of polar metabolites were analyzed using a single mobile phase and a Cogent Diamond HPLC column. The highly reproducible ANP method for hydrophilic compounds now enables the use of retention time as an orthogonal physical parameter for identification with an Accurate Mass Retention Time (AMRT) database. Work on creating the AMRT data base for hydrophilic compounds will be the focus of future wor.
