To study the role of superoxydase coenzyme in regulating Fas/FasL signal transduction and apoptosis of alveolar macrophages(AMs) in pneumoconiosis patients, The AMs of 50 male cases conducted massive whole lung lavage were collected and purified and divided into three groups (untreated group, Fas/FasL group and SOD group). AMs treated with SOD coenzyme or other block reagents for 24 hours in CO_2 incubator. SDS-page and Western-blot were used to detect Fas, FasL, Caspase-8 and Caspase-3. TUNEL and DNA fragmentation analysis were used to analyze AMs apoptosis. The apoptotic AMs index of SOD group (9.50%) and Fas/FasL group (14.01%) was lower than that of untreated one (19.18%). The DNA ladder pattern appeared in untreated group, but loomingly in Fas/FasL group and no ladder in SOD group. The expression of Fas, FasL, Caspase-8 and Caspase-3 in SOD group was lower than that of in the other two groups. SOD coenzymes can effectively regulate Fas/FasL signal transduction and block AMs apoptosis.
