Fluorescent labeled oligonucleotides had been extensively used for nucleic acid structure and dynamics studies as well as for diagnostic purposes such as single nucleotide polymorphism (SNP) detection etc. Several fluorescent bases have been used as site specific probe of nucleic acid structure and dynamics such as 2-aminopurine, 1,N~6-ethenoadenosine, N~2,3-ethenoguanosine, 3,N~4-ethenocytosine etc. However, it is mostly the fluorescence emission region that had been exploited for these studies. Fluorescence excitation band at 250-280 nm region of a fluorescently labeled oligonucleotide which is due to the singlet to singlet energy transfer from the fluorophore to the neighboring bases is not much utilized for structural studies or for diagnostic purposes. Here we present studies on the fluorescent excitation energy transfer region with N~1-wyosine incorporated deoxyoligonucleotides.
