1. SWATH data independent acquisition techniques are suitable for the identification of lipid peroxidation products and metabolites of polyunsaturated fatty acids. 2. Lipid peroxidation profiles in acute hepatic ischaemia-reperfusion injury have been determined using data independent acquisition and targeted MRM techniques. 3. Liver ischaemia-reperfusion injury results in significant sustained in vivo elevation in isoprostane E_2, which persists in the ischaemic reperfused lobe for at least 28 days after clamping, while F_2 isoprostane levels remain unaltered. 4. E_2 and F_2 isoprostanes are formed competitively. Depletion of cellular reducing agents favours the formation of E_2 over F_2 isoprostanes.
