-Negative ion mode MALDI IMS protocols were used to locate and identify phospholipids, fatty acids and flavonoids in Colombian coffee leaves. These compounds are important plant metabolites related to activation defense mechanisms in plants. -Detection of LMWM in MALDI IMS of coffee leaves is matrix-dependent. The use of DMAN, as MALDI matrix, allowed observation of signals corresponding to deprotonated molecules of fatty acids. When 1,5-DAN was used, signals corresponding to deprotonated molecules of flavonoids, phosphatidylglicerols (PG), phosphatidic acid (PA) and phosphatidylinositol (PI) were detected. Finally, 9-AA signals corresponding to deprotonated molecules of flavonoids, fatty acid and phosphatidic acid (PA). -LMWM have distinctive distributions on the leaves surface. While high abundance of fatty acids and flavonoids was observed on the leaves veins and margin, phosphatidylglicerols (PG), phosphatidic acid and phosphatidylinositol were uniformly distributed on the leaves.
