Screening Glycolipids Against Proteins in vivo using Picodiscs and Electrospray Ionization Mass Spectrometry

摘要

1) Picodiscs can be used to present glycolipids in a lipid environment for studying protein-glycolipid interactions in vitro. 2) The picodisc/CaR-ESI-MS method can detect both low and high affinity protein - glycolipid Interactions 3) The assay can be applied to picodiscs composed unknown lipid extracts, therefore, holds tremendous potential as a tool for the discovery of native receptors. 4) A comparison of the present results with data obtained with the CaR-ESI-MS assay implemented using NDs revealed that the PDs exhibit similar or superior performance to NDs for protein-GL binding measurement.
