Identification of a protein substrate specific for histone deacetylase 11 (HDAC11)

Our quantitative MS approach shows for the first time that histone HDAC11 functions as specific deacetylase of NPM1. Despite the low degree of acetylation of 1:100 as quantified by MS, NPM1 protein content and NPM1 degree of acetylation at Lys267 can both be quantified in total cell lysates with a single MS run using AQUA technology and MS1 analysis of the exact peptide masses. In summary, NPM1 acetylation at Lys267 can be assayed as a marker for HDAC11 activity.
