The demineralization fractions of bone protein extractions are often discarded in proteomic studies because they contains far less protein than the solubilization fractions (see Figure 1). Despite this disparity, our data show the demineralization fractions provide a much higher diversity of proteins and better coverage of bone/serum proteins in MS than those retrieved from an equal aliquot of protein from any subsequent solubilizing reagent.
