The aim of the present study was to develop a practical method for rapid identification and conformation of active constituents and metabolites in DanHong Injection by liquid chromatography-mass spectrometry (LC/MS/MS). Identification and structural determination of active constituents and metabolites is very important to evaluate the behavior of DanHong Injection in vivo study. Mass spectrometric data were acquired in the positive and negative mode. Information Dependent Acquisition (IDA) workflows, including EMS-IDA-EPI, Pre/NL-IDA-EPI and MIM-IDA-EPI, were designed for the rapid identification of active compounds in DanHong Injection. pMRM-IDA-EPI was used to acquire data for the detection of metabolites in rat serum on a hybrid triple quadrupole-linear ion trap mass spectrometer. A total of 45 compounds were identified and confirmed using SCIEX QTRAP 4500 system coupled to liquid chromatography, including 32 major compounds of Dandong injection and 13 metabolites in rat serum samples.
