Fast Analysis of Therapeutic Monoclonal Antibodies, Fragments, and Oxidation Variants Using a Super-Macro Porous Reversed-Phase Column Coupled with an Orbitrap Mass Spectrometer

摘要

mAb LC, HC, Fc, Fab, scFc and F(ab')_2 fragments are successfully separated using MAbPac RP column and a 10-min gradient. Mass of oxidized HC is fully resolved from non-oxidized HC using Q Exactive Plus mass spectrometer. Separation of oxidized versus non-oxidized fragments is improved using 0.1% TFA-containing mobile phases.
