摘要

SICAP is a powerful method for comprehensive and dynamic chromatin profiling The procedure starts after cell fixation, precluding perturbation of biological processes. SICAP includes a proper negative control to exclude contaminations, requires 2-4 million cells, and is compatible with the analysis of tissues. SICAP identified about 3000-4000 proteins in chromatin; this number is in line with other recent studies. SICAP has demonstrated the ability to identify novel chromatin components, such as secreted factors and metabolic enzymes. Direct comparison to full proteome analysis demonstrates extensive protein translocation to/from chromatin depending on the pluripotent state.
