Multiplexed Protein Expression Profiling of Pancreatic Stellate Cells under Nicotine Stress

Various nicotinic receptors are expressed in pancreatic stellate cells (PaSC). Nicotine increases migration and cell proliferation of PaSC. Over 6,000 proteins were quantified per cell line via TMT-MS3 analysis. Of these, we determined 151 proteins as being differentially abundant with statistical significance (Benjamini-Hochberg corrected p-value <0.01) and fold changes >±1.5. Proteins determined to be of lower abundance upon nicotine treatment include those of proteasomal, and ribosomal origin. Conversely, proteins of higher abundance include immunomodulating proteins, members of complement system, SLC transporters, and some nAChR-binding proteins (apolipoprotein A-I and amyloid beta).
