Backflush and deconvolution are discussed and applied to the urinary organic acid analysis. Backflush removes the high boilers quickly and at a lower oven temperature, therefore, improves column lifetime and cycle time. The introduction of deconvolution to the analysis of urinary organic acids can provide two very significant advances in this important area of metabolic disease diagnosis: (i) DRS with an application-specific library is able to identify co-eluting compounds including those with similar spectra such as Citric acid-4TMS and Isocitric acid-4TMS. This identification (by library search) was not possible without DRS. (ii) The quant result comparison shows some of the significant advantages of DRS over the standard ChemStation or metabolic lab protocol. Deconvolution helps to find and quantify all target compounds accurately. 2X speed-up method reduces half of run time and still obtain excellent semi-quantitation results.
