Several peptides from various dehydrogenases were identified in the preparation. We are currently studying their homologies to understand the minimum similiarity required for activity. We found that B cells contain 5-HEDH, and since they also express 5-lipoxygenase, they can synthesize 5-oxo-ETE from arachidonic acid in the presence of calcium ionophore. We are also further purifying 5-HEDH by affinity chromatography on stationary phases containing bound substrate analogs. This should greatly increase the purity of 5-HEDH, making it easier to identify potential candidates by mass spectrometry. 5-HEDH purified from human neutrophils, will also be analyzed by mass spectrometry.
