Simultaneous LC-ESI-MS-MS Determination of Cyclosporine A, Tacrolimus, and Sirolimus in Blood and Mycophenolic Acid in Plasma, Using Common Pretreatment Procedure.

The purpose of the study was to develop rapid and simple procedure for simultaneous determination of cyclosporin A (Cs A), tacrolimus (T), and sirolimus (S), mycophenolic acid (MPA) as well as ascomycin, cyclosporin D , and desmethoxysirolimus used as internal standards, using common pretreatment procedure. In the method development, six-level blood calibrators and four level QC standards were used for Cs A, S, and T. Two level QC plasma standards were used for MPA, together with four-level calibrators. All QC standards and calibrators were supplied by Chromsystems Instruments and Chemicals, Munich, Germany, only calibrators for MPA were perepared in the lab.
