The application of LC-MS/MS for rapid compound profiling and characterization is of increasing interest in drug discovery and development process. Traditionally, selected reaction monitoring (SRM or MRM) has been a major method for drug metabolites identification. However, this experiment misses information on unpredicted metabolites. Other approaches including precursor ion and neutral loss scanning have to be employed resulting in multiple LC-MS/MS experiments. The combination of such experiments is both time and sample consuming. In this work we described a rapid method for drug metabolite analysis using Finnigan LXQ linear ion trap mass spectrometer. With this approach, a single LC-MS/MS run is sufficient for comprehensive drug metabolites profiling and identification, enabling high throughput metabolism studies.
