We examined whether disruption of ascorbic acid (AA) status contributes to oxidative damage in the liver of rats with water immersion restraint stress (WIRS). Wistar rats were subjected to WIRS at 23degC for 1.5, 3 or 6 h. Liver damage appeared at 15 h of WIRS and developed at 3 and 6 h. The liver of rats with 3 or 6 h of WIRS had decreased total AA, reduced AA, and reduced glutathione contents and increased oxidized AA content but showed no change in vitamin E content. Hepatic lipid peroxide content increased at 3 and 6 h of WIRS. When AA (500 mg/kg) was orally administered to rats subjected to WIRS just before the onset of the stress, all these changes found at 3 and 6 h of the stress were attenuated almost completely. These results indicate that disruption of AA status contributes to oxidative damage in the liver of rats with WIRS.
