Implant materials, when inserted into the body, interact with the internal environment and can be markedly changed by this interaction. Our laboratory has previously shown that hydroxyapatite (HA), a calcium phosphate type of biomaterial, is very efficient at adsorbing proteins from blood that promote the adhesion and spreading of mesenchymal stem cells (MSCs) [1], an osteoprogenitor cell type. In fact, human MSCs adhere better to HA coated with fetal bovine serum (FBS), than to HA coated with purified fibronectin, vitronectin or RGD peptides [2]. These data call into question whether functionalizing HA with proadhesive peptides/proteins would have a therapeutic benefit, given that HA implants are coated with the patient's blood upon insertion. In an ongoing effort to understand, and ultimately optimize, events occurring at the tissue/implant interface, our laboratory has, in the present study, evaluated the adhesion and spreading of human MSCs on HA and tissue culture plastic (TCP) coated with either FBS or human serum (HuS). In addition, we have analyzed cell behavior on collagen-coated biomaterials, given that the organic phase of bone matrix is composed primarily of collagen I. The key findings of this study are that: 1) cells behave differently on serum-coated HA than on serum-coated TCP, most probably due to differences in the profile of proteins that adsorb to these diverse substrates and 2) collagen I is equivalent to, or better than, whole serum in stimulating cell adhesion to HA and TCP, implicating a role for this matrix molecule in promoting MSC/biomaterial interactions.
