CONSTRUCTION OF MORE REFINED COMPARATIVE MAPS FOR PIG CHROMOSOMES 9 AND 10

To identify positional candidate genes tor quantitative trait loci (QTL) on pig chromosomes (SSC) 9 and 10, more refined comparative maps have been constructed. The comparative maps of SSC9 and SSC 10 were originally made by heterologous chromosome painting and provide a low resolution view of evolutionary breaks that have occurred throughout pig and human chromosomal evolution. These maps have revealed that sections of SSC 10 are homologous to regions of human chromosomes (HSA) 1, 9 and 10, and thatSSC9 is homologous to HSA1, 7 and 11. However the boundaries of these regions are not clearly defined. Relevant chromosomal regions of the public human genome sequence were used in a BLAST of pig expressed sequence tag (EST) database. Matching ESTs wereused to design 49 pig specific primers. Genes were then assigned to pig chromosome using the INRA-somatic cell hybrid panel (INRA-SCHP) and high resolution radiation hybrid panel (IMpRH). Seventeen genes were mapped to SSC9, and sixteen to SSC 10 as predicted. However, eleven genes were localized to SSC1, three to SSC3 and three to SSC4. The physical mapping of these genes has contributed to refinement of the comparative maps, and has also improved the identification of evolutionary breakpoints betweenpigs and humans.
