High DNA Fragmentation Indices on the Sperm Chromatin Structure Assay Correlate More with Increased Risk of Miscarriage Rather than Conception Failure in Women Who Were Refractory to Achieving Successful Pregnancy Following

Initial studies of the sperm structure chromatin assay suggests that high DNA fragmentation indices (DFT) (>30%) were associated with minimal chance for pregnancy. The present study was started when these initial data were reported. The couples evaluated had to have a minimum of 2 previous failures to successfully deliver babies from in vitro fertilization-embryo transfer (IVF-ET) to be eligible for the study. DFI scores >30% were found in 29 male partners. The clinical pregnancy rate (PR) was 27.6% (8/29) and the miscarriage rate was 62.5% (3/8). There were 41 males with a DFI score of 15-30 with a clinical PR of 31.7% (13/41) (p=NS). The miscarriage rate was 38.4% (5/13). A normal DFI was found in 30 males with a clinical PR of 36.1% (13/38). The miscarriage rate was 46.1% (6/13).
