Highly Efficient Automated Synthesis of ~11C-Choline

Introduction [~11C]-Choline has been reported a potential tracer for imaging a variety of human tumors with positron emission tomography (PET). Materials and Methods [~11C]-CH_3I was bubbled into the reaction vessel of the synthesizer filled with 2mg DMAE in 50muL acetone at 0°C, instead of 500muLDMAE of 60muL on C-18. The [~11C]-Choline was puritied on Sep-Pak CM column. Results The synthesis yield was improved from 82.0% to 96.5%(EOB), while the precursor DMEA decreased from 60mg to 2mg. Absolute yields of [~11C]-Choline was 2500MBq at a 10min irradiation of 15muA, and a total synthesis time less than 8min from [~11C]-CH_3I to [~11C]-Choline. Conclusions A highly efficient synthesis route for the production of [~11C]-Choline in an automated synthesizer was developed.
