Mammalian fuel metabolism, even within a single tissue, is made up of complex,over-lapping, convoluted and redundant pathways. These are punctuated by futilecycles, sequestered within quasi-distinct substrate pools, and influenced by amultitude of effectors. This complexity is augmented by the existence, in the samecellular milieu, of the signaling pathways that regulate metabolic flux. Because of thiscomplexity the ability to determine the role of specific enzymes is a nearoverwhelming challenge. Undaunted, we endeavor to investigate the metabolic,enzymatic, and signaling mechanisms that work to orchestrate these pathwaysbecause, by elucidating these, we may begin to understand devastating metabolicdiseases, such as obesity and diabetes.
