Development of Micro Chemical Reaction Devices with Ultra-low Volume Liquid Manipulation

We have developed novel micro devices for chemical reactions in which nanoliter-sized multiple droplets can be accurately metered and mixed. These devices consist of three-dimensional microchannel networks fabricated on polydimethyl- siloxane (PDMS), a kind of silicon elastomer. Hydrophobic and narrow channels called 'hydrophobic passive valves' act as a kind of valve, and help accurate liquid manipulation. With this method, single reagent injection is enough to form multiple nanoliter-sized aliquots for multiple different reactions. We designed microdevices for 3 and 50 reactions, and evaluated the ability and reproducibility, which proved to have sufficient reliability for quantitative reactions. For the application, we performed biochemical analysis using enzymatic reaction. This liquid manipulation method will be a useful tool in the field of miniaturized chemical reaction systems by integrating with other devices such as micro optical or electrical sensors.
