Here we describe flowcytometric (FCM) analysis of HOS 58 cells, a model cell line of human osteoblasts, by using human bone specific alkaline phosphatase (ALP) monoclonal antibody. After the cells at culture day 7 were incubated with FITC labeled human bone specific ALP monoclonal antibody, the cells were analyzed by FCM. We used monoclonal antibodies derived from two different hybridomas. Both antibodies recognize the cells with high ALP activity, especially about 80% of the total cells incubated with antibody from hybridoma B4-78 showed high fluorescent intensity Based on the facts that the composites of cultured osteoblasts and porous ceramics can show immediate in vivo bone forming capability, present results suggest that this isolation and enrichment technique of osteoblasts can be available in bone reconstruction surgery.
