Sulfolobus solfataricus carboxypeptidase (CPSso), is a heat- and pressure resistant metalloprotease. Thanks to its properties it is an ideal tool for investigations aimed at understanding the role of non-covalent interactions in substrate binding. Actually, its catalytic and kinetic mechanisms are well understood; also, it can cleave almost any N-blocked amino acid, and the hydrolytic reaction is easily detectable. In addition, the meaning of k_(cat) and K_m in terms of rate constants is well understood. Thanks to these properties, we could dissect, to some extent, the individual non-covalent interactions involved in substrate binding by determining the kinetic parameters as a function of pressure and temperature.
