INTRAOSSEOUS DELIVERY OF rhBMP-2/ HYALURONAN FOR OSTEOPOROSIS

rhBMP-2 was delivered from hyaluronan based materials formulated into solid depot rods for local intraosseous delivery to induce new bone formation. By varying dose and excipients, in vitro rhBMP-2 release was modulated to exhibit slow sustained release to fast burst release in bioactive form. In one formulation, in vivo release kinetics of the ~(125)I-rhBMP-2 / hyaluronan rods in rabbit distal femur intraosseous site revealed that rhBMP-2 was released to the local bone for over 10 weeks. Rat subcutaneous implants of the rhBMP-2 rods revealed bone formation in the surrounding tissue. These results indicate that local intraossesous delivery of rhBMP-2 may be a promising therapy for treatment of osteoporosis.
