One of the most characteristic properties of proteins is the elliptical outline in the stability diagram. Hereby, it is not only possible to denature the protein by application of high temperature but also by using high pressure or a combination of both. The elliptical outline of the phase diagram can be found by following either the rate of inactivation of the protein under different conditions or by plotting the transition pressure and temperatures of denaturation. In this study, we followed the rate of hydrogen- deuterium exchange in lipoxygenase. The aim was to establish a correlation in the phase diagram with previous measurements based on inactivation and denaturation. It is shown that by following kinetic parameters, one can estimate the stability of the protein under different conditions.
