Investigations were carried out to establish a reliable in vitro culture system for high frequency and to study regeneration via somatic embryogenesis and biochemical aspects involved in the pathway by using mature seed explants of indica rice cultivar CO 45, The best callus induction response was obtained on MS medium supplemented with 2,4-D, kinetin, and sucrose. A medium good for germination of somatic embryos and regeneration was also identified, Histological studies indicated evidence for the pathway of regeneration via somatic embryogenesis. Protein analysis through SDS-PAGE revealed distinguishable differences in protein profiles among the different stages of embryogenesis. Markers for embryogenic calli and for characterizing germinating somatic embryos were identified. Progenies of regenerants showed variation for economic attributes.
