Current methods of evaluating leukocytes in birds are relatively subjective and limited to the use of either the Eosinophilic Pippette system or the Natt and Herrick method, neither of which are as precise as automated methods. None evaluate leukocytefunction. Confounding the process of differentiation is the difficulty in distinguishing some lymphocytes from monocytes. Using a fluorescein (FITC)-labeled opsonized E coli bacteria and flow cytometry, lymphocytes can be distinguished from monocytes and the phagocytic activity of monocytes and granulocytes can be quantified and evaluated.
