Cloning and analysis of the gene from Xanthomonas citri involved in plant growth

Periplasmic glucans has been suspected to be responsible for the pathogen to attach to the host plant cell especially in the cause of the infection by Agrobacterium tumefaciens and Pseudomonas syringae. The pathogen of citrus canker, Xanthomonas citri, produces a very unique periplasmic glucan. We have isolated the transposon Tn3-Spice mutant inserted into the region homologous to chvA of A. tumefaciens from X.citri, and this mutant was shown to be deficient in multiplication of the pathogen in the host plant. Here, we report the cloning and analysis of the ORF where Tn3-Spice was inserted, as it should be involved in in planta growth of the pathogen. From homology search, this gene was suggested to encode for the secretory function during the first stage of its infection.
