Recent developments in oral vaccination systems focused on the interest of biodegradable microspheres. So, prior to invivo targeting studies, the use of an ex-vivo model, to characterize their potentialities, based on particles uptake is an important tool. Phagocytosis study has been essentially focused to understand specific aspects of phagocytosis process in host defense. Among the methods, microscopic study has often been used, but according to our previous works based on culture plates, this method presented drawbacks: small proportion of the population can be evaluated and hetergeneous distribution of particles was observed. In order to avoid these problems, another model has been based on suspension cells culture with a flow cytometry evaluation.
