Recurrent luminal narrowing (restenosis) as a result of excessive vascular smooth muscle cell (VSMC) proliferation remains a principal limiting factor for the long-term success of vascular surgery. It has been known that in order to inhibit VSMC proliferation drugs should be delivered at high concentration for an extended period of time. Therefore, a nondiffusible particulate micro carrier system has been applied for a vascular retentive sustained drug delivery system. However, microparticles showed poor efficiency of arterial uptake and possibility of causing ischemia due to clogging blood capillaries. Therefore, this study was undertaken to formulate Genexol~(TM) (PT, plant cell cultured paclitaxel, Sam Yang Genex Inc.), an antimicrotubule agent, into biodegradable poly(ethylene oxide)-poly(lactide/glycolide) (PEO-PLGA) nanospheres and study their effect on VSMC proliferation in culture.
