The establishment of avian embryonic culture is important for both analysis of developmental process and establishment of transgenic chickens which produce useful biological materials in egg contents. However, hatchability of cultured embryos was at most 50percent by now. We identified that the decrease in hatchability for cultured embryos was caused by limitation of oxygen and calcium supplies. In quail embryo culture using a chicken egg shell as a culture vessel, viability in middle phase of culture was improved and 30percent of embryos hatched by oxygen enrichment. Furthermore, hatchability was improved very much (80percent) by the addition of Ca lactate in addition to oxygen aeration.
