A novel isoflavone, 7, 2', 4'-trihydroxyisoflavone-4'-O-beta-D-glucopyranoside was isolated from the aerial part of Crotalaria sessiliflora. The isoflavone glucoside was found to enhance the proliferation of the MCF-7 human breast cancer cell line, which possesses estrogen receptors (ERs) and responds in culture to estrogen. The estrogenic property of the isoflavone glucoside was blocked by the known ER antagonist tamoxifen, indicating the involvement of the ER. Furthermore, the isoflavone was foundto enhance the acetylcholinesterase (AChE) activity of the rat neuronal cell line PC12 treated with a low concentration (0.1 muM) of nerve growth factor (NGF). In this study, it was shown that the phytoestrogen enhances AChE activity in PC 12 cells by binding to the ER; the actual mechanism will be determined by proteome analysis.
