The objectives of this pilot study were to evaluate a recently developed neutral endopeptidase inhibitor as a possible treatment for congestive heart failure in dogs.More specifically, the study was designed to evaluate the clinical, hemodynamic,neuroendocrine, and renal effects of orally administered ecadotril (BAY y 7432) in dogs with myocardial failure induced by rapid ventricular pacing.The evaluation of this compound included assessements of cardiac structure and function and its effect on exercise capacity and survival. Ecadotril is the S-enantiomer of the racemate, acetorphan, and its chemical name is N-(S)-[2-[(Acetylthio)methy1]-1-oxo-3-phenyl propy1]-glycine benzylester. Ecadotril,formerly known as sinorphan, is a pro-drug that is metabolized to the active compound S-thiorphan, a potent inhibitor of the enzyme neutral endopeptidase.
