In natural aquatic environments and biological wastewater treatment systems, eukaryotic microorganisms such as protozoa and rotifer are very important in term of predator for controlling balance of the food web in the ecosystem. Therefore, isolation, identification and pure cultures of such predation microorganisms had to be conducted as the basic study to understand aquatic ecosystem or to develop the eco-friendly remediation method for water environments. However, morphological identification and the culture of such microorganisms demand higher skill. Moreover it is impossible to culture many microorganisms in laboratory. Now a day, molecular ecological methods have been applied for the study of eukaryotic microorganisms. However, the gene database is not enough for eukaryotic microorganisms. Thus we tried to one individual and one cell PCR for eukaryotic microorganisms in activated sludge system and environments toward the investigation for non-culturable eukaryotic microorganisms.
