Histological Observation of Vitreous Cryopreservation of Tissue Engineered Tendon Implanted in Rats

The purpose of this paper is to investigate the effects of the vitreous cryopreservation of tissue engineered tendon implanted in vivo on repairing the tendon defects of the rats. Sixty-four adult SD rats were randomly divided into two groups, vitreous cryopreservation tissue engineered tendons and fresh tissue engineered tendons without cryopreservation were implanted respectively. The 5 mm tendon defect models were prepared in the middle part of Achilles tendons of the rats, the tissue engineered tendons were implanted for repairing the defects of tendons. In the 2nd, the 4th , the 6th and the 8th weeks after surgery, the general morpha and histological change of the implanted tendons and surrounding tissues were observed. The results show that no significant differences in general morpha and histological change of the implanted tendons and surrounding tissues were observed between two groups. The implanted tendon materials were degraded gradually and were infiltrated and surrounded by a large number of inflammatory cells and proliferous fibrous connective tissues. In the 6th and the 8th weeks after the surgery, most parts of the implanted tendons were degraded, absorbed and replaced by a large number of regenerated collagenous fiber tissues. The morphology and orientation were similar to those of normal tendon tissues. Collagen fibers were connected well in bridge site of the tendons, and the tendon defects of the rats were repaired basically. The results indicate the vitreous cryopreservation tissue engineered tendons implanted in vivo can repair the tendon defects of the rats.
