Antioxidant Activities and Purification of Polysaccharide Extracted from the Fruiting Bodies of Phellinus Igniarius

Objective: To study the vitro antioxidant activity and structure of polysaccharide extracted from the fruiting bodies of Phellinus Igniarius used different extraction ways. Method: Used ultrasound assisted cellulase enzymes, hot-water extraction to extract crude polysaccharide (PIPs) and protein remove by sevage method. The inhibition effect of polysacchatide on superoxide anion radical from the pyrogallol autoxidation reaction and hydroxyl radical from Fenton reaction was measured. M80 and C80, which have good antioxidant activity, were isolated by DEAE-Sephadex A-52 column chromatography, then purified by Sephadex G-100 column chromatography. Infrared spectrometry analyzed the structural differences between W80 and C80. Results: PIPs had different degrees of clear roles on superoxide radical and hydroxyl radicals, the effect of W80 and C80 were more obvious. Infrared spectroscopy exhibited that the major functional groups of W80 was almost consistent with C80. Conclusion: Antioxidant activity and the structure of polysaccharides may not be damaged by combinatorial treatments of ultrasonic waves and cellulase enzymes.
