Adverse skin irritation reactions often limit the number of potential drug candidates for transdermal drug administration. We studied the irritation potential of tacrine (a drug to treat Alzheimer's disease), iontophoresis and their combination in human volunteers. The adverse reactions experienced included pinching-sensations, erythema, drying of the skin and coldness on the skin and fingertips. Skin irritation reactions were correlated directly to the current density used. None of the adverse effects were regarded serious. No additional damage on the skin could be attributed to the use of tacrine in these short experiments.
