The effects of glycerin on stratum corneum and dermis were evaluated by measuring the skin permeation rates of nicotinic acid (NA) with freshly excised hairless mouse skin. The steady state skin permeation rates of different concentrations of glycerin either in donor or receptor solutions were determined and the relationship between the degree of hydration and the skin permeation rate was investigated. Glycerin in receptor solution facing dermis side significantly influenced the skin permeation rate of NA and the enhancement was concentration dependent. The steady state skin permeation rate showed the highest in 10% and lowest in 50% (19.25 vs 2.59 D g/cm~2/hr). The enhancement effect of glycerin in donor solution showed similar profile but less significantly ( 6.39 vs 2.71 D/cm~2/hr). The effect of hydrating vehicles in receptor fluids on the skin permeation rate should be carefully considered.
