A new in situ experimental method to evaluate in vivo skin disposition of drug after topical application in hairless rats

Topical formulations on skin can be classified into two categories according to the target site of action of the containing drugs; those for the systemic action and those for local (cutaneous tissues) effect. The pharmacological effects especially for the latter formulations are greatly influenced by the cutaneous disposition of drugs after topical application. Analysis of the drug disposition kinetics in skin, therefore, is important to assess such formulations. However, no sufficient explanation has been elucidated yet whether the topical effects are due to direct penetration or secondary distribution of drugs via the local vasculature following the systemic absorption.
