A novel concept of combination chemotherapy and photodynamic therapy (PDT) using N-(2-hydroxypropyl)methacrylamide (HPMA) copolymer bound drugs was recently proposed. On two cancer models, Neuro 2A neuroblastoma and human ovarian carcinoma heterotransplanted in nude mouse it was shown that combination theapy with HPMA copolymer bound anticancer drugs, meso-chlorin e_6 monoethylenediamine (MCe_6) and adriamycin (ADR), produced tumor cures which could not be obtained with either therapy alone. The efficacy of combination chemotherapy and PDT depends on the time lag between the administration of both conjugates and irradiation (to activate the photosensitizer, Mce_6). To optimize the treatment protocol, the biodistribution of free and HPMA copolymerbound Mce_6 and ADR in nuu athymic mice were studied.
