We synthesized a series of differentially branched ester amine quadpolymers (BEAQs) to assess the influence of branching and polymer structure function relationships with gene delivery efficacy and specific cellular level barriers to transfection such as lysosome avoidance and nuclear entry. The optimal BEAQs demonstrated marked improvement in delivery of plasmid DNA over optimized previous generation PBAEs. attributable to increases in secondary amine content and endosomal escape at lower equivalent polymer masses.
